Peer-Reviewed Research

Semax is a synthetic heptapeptide derived from adrenocorticotropic hormone (ACTH 4–10). Current research has investigated Semax in laboratory, animal, and translational neuroscience models involving neuroprotection, ischemic stroke, traumatic brain injury, oxidative stress, Alzheimer's disease, spinal cord injury, and neuroinflammation. Although Semax has a longer clinical history in Russia, relatively few recent international human clinical trials have been published. Much of the current literature remains preclinical. Semax peptide targets the μ opioid receptor gene Oprm1 to promote functional recovery after spinal cord injury

Journal: British Journal of Pharmacology
Published: 2025
Study Type: Original preclinical research

Summary

Researchers evaluated Semax in a mouse model of spinal cord injury. Treatment improved neurological recovery, reduced neuronal damage, and identified Oprm1 as a potential molecular target associated with functional recovery. The findings support continued investigation of Semax in experimental neuroregeneration but do not establish efficacy in humans.

Semax decreases copper-catalyzed reactive oxygen species production and amyloid-β cytotoxicity

Journal: Bioinorganic Chemistry and Applications
Published: 2025
Study Type: Original laboratory research

Summary

This laboratory investigation evaluated Semax in Alzheimer's disease–related experimental models. Researchers reported reductions in copper-mediated oxidative stress and decreased amyloid-β cytotoxicity through mechanisms involving metal-ion interactions. The study provides mechanistic insight into Semax's neuroprotective activity but was not performed in human participants.

The effect of Semax on intracellular calcium dynamics in rat brain neurons

Journal: Bulletin of Experimental Biology and Medicine
Published: 2025
Study Type: Original laboratory research

Summary

Researchers investigated how Semax influenced intracellular calcium signaling in rat hippocampal neurons. The findings demonstrated changes in calcium dynamics associated with neuronal signaling pathways involved in cellular function and neuroprotection. Additional studies are required to determine the clinical significance of these findings.

Semax affects copper-induced amyloid-β aggregation in artificial membrane models

Journal: ACS Chemical Neuroscience
Published: 2022 (Landmark Study)
Study Type: Original laboratory research

Summary

This mechanistic study demonstrated that Semax reduced copper-induced amyloid-β aggregation and associated cytotoxicity in experimental membrane models. The findings remain an important reference for understanding Semax's proposed neuroprotective mechanisms.
